Mechanism of Action
Macitentan blocks endothelin-1, a potent vessel-constricting peptide, from binding to ETA and ETB receptors in pulmonary blood vessels. This reduces vasoconstriction and abnormal vessel-wall thickening, lowering pulmonary vascular resistance.
