Mechanism of Action
Methoxsalen is a furocoumarin that intercalates between DNA base pairs. On UVA exposure (320–400 nm), it forms covalent cross-links with thymine on opposite DNA strands, halting DNA synthesis in dividing cells. In psoriasis this suppresses hyperproliferative keratinocytes; in vitiligo it stimulates melanocyte migration from hair follicles into depigmented patches.
Dosage & Administration
- Dose by body weight: 0.6–0.8 mg/kg (typically 1–3 tablets) 1.5–2 hours before UVA
- Take with food to reduce GI upset
- Use protective UV eyewear 24 hours after dose
