Mechanism of Action
Amantadine enhances dopaminergic transmission by promoting presynaptic dopamine release, blocking dopamine reuptake, and weakly stimulating dopamine receptors. It is a non-competitive antagonist of NMDA glutamate receptors, which underlies its antidyskinetic effect by modulating overactive corticostriatal glutamatergic signalling.
Dosage & Administration

- Elderly and renally impaired patients: 100 mg once daily or less.
- Avoid late-evening doses (may cause insomnia).

Side Effects
- Insomnia, nervousness, dizziness, peripheral oedema, livedo reticularis (mottled skin discolouration), dry mouth, blurred vision, constipation, urinary retention.
- Rarely: confusion, hallucinations (more common in elderly and renal impairment), seizures.

- Hypersensitivity to amantadine.
- Untreated narrow-angle glaucoma.
- Severe renal impairment.
- History of seizure disorder requires caution.
