About Makclav-150 Injection
Makclav-150 Injection provides Amoxicillin 125 mg + Clavulanic Acid 25 mg — paediatric/neonatal IV strength for hospitalised infants requiring weight-appropriate IV beta-lactam therapy.
Manufactured by Trumac Healthcare under WHO-GMP. Available for monopoly-based PCD pharma franchise and third-party manufacturing.
Indications
- Neonatal sepsis (with appropriate weight-based dosing)
- Paediatric severe pneumonia
- Paediatric pyelonephritis
- Neonatal meningitis (selected)
Mechanism of Action
Amoxicillin is a beta-lactam (aminopenicillin) that binds penicillin-binding proteins (PBPs) inhibiting bacterial cell wall peptidoglycan cross-linking. Clavulanic Acid is a beta-lactamase inhibitor — irreversibly inactivates bacterial beta-lactamases protecting Amoxicillin from enzymatic hydrolysis. The combination extends Amoxicillin's spectrum to include beta-lactamase-producing strains of E. coli, H. influenzae, M. catarrhalis, and many anaerobes.
Dosage & Administration
Infants/children: 25–45 mg/kg/day Amoxicillin component IV divided 8-hourly.
Side Effects
Diarrhoea (more common than amoxicillin alone — Clavulanate-related), GI upset, rash, candidiasis. Cholestatic hepatitis (rare). C. difficile colitis.
Contraindications & Precautions
Penicillin/cephalosporin hypersensitivity, history of Amoxicillin-Clavulanate-induced jaundice or hepatic dysfunction, infectious mononucleosis (rash), severe renal impairment (dose adjust).
Storage
Store 2–25 °C. Use immediately after reconstitution.
