About Glimstay-MP2 SR Tablet
Glimstay-MP2 SR is the triple-mechanism combination — Glimepiride for insulin secretion, Metformin for insulin sensitivity, and Pioglitazone for additional peripheral insulin sensitisation. For Type 2 diabetes inadequately controlled on dual therapy.
Pioglitazone activates PPAR-gamma receptors, improving peripheral insulin sensitivity at adipose and muscle level.

Mechanism of Action
Glimepiride is a third-generation sulphonylurea — longer duration of action, lower risk of severe hypoglycaemia than older agents, and weight-neutral compared with insulin secretagogues. Metformin reduces hepatic glucose production (gluconeogenesis), improves peripheral insulin sensitivity, and reduces intestinal glucose absorption — without causing hypoglycaemia in monotherapy. Pioglitazone activates PPAR-gamma nuclear receptors enhancing adipose insulin sensitivity, reducing hepatic glucose production and improving peripheral glucose uptake.
Dosage & Administration
Adults: 1 tablet once daily with breakfast.
Side Effects
Hypoglycaemia, weight gain (less with Glimepiride), nausea, mild GI upset. Rare hyponatraemia. Weight gain, peripheral oedema, fracture risk (Pioglitazone). Bladder cancer signal (controversial).
Contraindications & Precautions
Type 1 diabetes, diabetic ketoacidosis, severe hepatic/renal impairment, pregnancy, lactation, sulpha allergy. Heart failure (NYHA III-IV), bladder cancer, macular oedema, severe osteoporosis.
