About Aciron-250 Injection
Aciron-250 Injection from Trumac Healthcare contains 250 mg of lyophilised Acyclovir, a synthetic purine nucleoside analogue with potent antiviral activity against Herpes Simplex Virus types 1 and 2, Varicella-Zoster Virus and, to a lesser extent, Epstein-Barr Virus and Cytomegalovirus.
Acyclovir is selectively phosphorylated by viral thymidine kinase to its active triphosphate form, which inhibits viral DNA polymerase and terminates DNA chain elongation. Because this activation step is virus-specific, Acyclovir has very low toxicity to uninfected host cells, making it the standard of care for severe and disseminated herpes infections.
The intravenous formulation is indicated when oral therapy is inappropriate — in immunocompromised patients, herpes encephalitis, neonatal HSV, severe varicella and disseminated zoster. Mechanism of Action
Acyclovir is phosphorylated to acyclovir monophosphate by viral thymidine kinase, then to acyclovir triphosphate by cellular kinases. Acyclovir triphosphate competitively inhibits viral DNA polymerase and is incorporated into the growing viral DNA strand, causing chain termination and arresting viral replication.
Dosage & Administration
Adults and children > 12 years: 5–10 mg/kg body weight every 8 hours by slow IV infusion over at least 1 hour, for 5–10 days depending on indication. Adjust dose in renal impairment. To be prescribed and administered under qualified medical supervision only.
Side Effects
Local irritation or phlebitis at the injection site, nausea, vomiting, headache, elevated serum creatinine, transient rise in liver enzymes. Rare: crystalluria, acute renal failure (reduced by adequate hydration), neurotoxicity, thrombocytopenia.
Contraindications & Precautions
Known hypersensitivity to acyclovir or valacyclovir. Use with caution in renal impairment, dehydration, the elderly, and in patients receiving other nephrotoxic agents. Adequate hydration is required to prevent crystalluria.
