Mechanism of Action
Domperidone is a peripheral D2 receptor antagonist — enhances upper GI motility, accelerates gastric emptying, increases LOS tone. Does not cross blood-brain barrier — no extrapyramidal effects at therapeutic doses. Dimethicone is anti-foaming agent reducing gas bubble surface tension.
Dosage & Administration
Infants: 2.5–5 ml before feeds.
Side Effects
Hyperprolactinaemia (galactorrhoea, gynaecomastia, amenorrhoea), QT prolongation (short-term high-dose), GI cramps. Mild diarrhoea.
Contraindications & Precautions
Prolactinoma, QT prolongation, on QT-prolonging drugs, severe hepatic impairment, GI obstruction/haemorrhage.
