Mechanism of Action
Sodium Valproate is a broad-spectrum AED — increases GABA levels (inhibits GABA transaminase), blocks voltage-gated sodium channels, and inhibits T-type calcium channels. Also used as mood stabiliser in bipolar disorder and migraine prophylaxis.
Dosage & Administration
Children: 15–30 mg/kg/day divided into 2–3 doses.
Side Effects
Tremor, weight gain, hair loss (alopecia), hepatotoxicity (severe in children <2 years), pancreatitis, thrombocytopenia, teratogenicity (neural tube defects). Severe hepatotoxicity in young children.
Contraindications & Precautions
Pregnancy (CATEGORY X — teratogenic), hepatic dysfunction, urea cycle disorders, mitochondrial disorders, children <2 years on polytherapy.
