About Makclav-ES Dry Syrup
Makclav-ES Dry Syrup provides Amoxicillin 600 mg + Clavulanic Acid 42.9 mg per 5 ml — the extra-strength (ES) ratio (14:1) optimised for penicillin-resistant pneumococci and resistant H. influenzae. Used for severe paediatric AOM, sinusitis, and recurrent pneumonia.

Mechanism of Action
Amoxicillin is a beta-lactam (aminopenicillin) that binds penicillin-binding proteins (PBPs) inhibiting bacterial cell wall peptidoglycan cross-linking. Clavulanic Acid is a beta-lactamase inhibitor — irreversibly inactivates bacterial beta-lactamases protecting Amoxicillin from enzymatic hydrolysis. The combination extends Amoxicillin's spectrum to include beta-lactamase-producing strains of E. coli, H. influenzae, M. catarrhalis, and many anaerobes. The high Amoxicillin:Clavulanate ratio (14:1) provides higher Amoxicillin tissue levels while minimising Clavulanate-related diarrhoea.
Dosage & Administration
Children: 90 mg/kg/day of Amoxicillin component divided every 12 hours.
Side Effects
Diarrhoea (more common than amoxicillin alone — Clavulanate-related), GI upset, rash, candidiasis. Cholestatic hepatitis (rare). C. difficile colitis.
Contraindications & Precautions
Penicillin/cephalosporin hypersensitivity, history of Amoxicillin-Clavulanate-induced jaundice or hepatic dysfunction, infectious mononucleosis (rash), severe renal impairment (dose adjust).
