Mechanism of Action
Orlistat irreversibly inhibits pancreatic and gastric lipases — preventing hydrolysis of dietary triglycerides into absorbable monoglycerides and fatty acids. Approximately 30% of dietary fat is excreted unchanged in faeces.
Dosage & Administration
- Adults: 120 mg three times daily with or up to 1 hour after each main meal containing fat.
Side Effects
- Oily spotting/stools (decrease with reduced fat intake), faecal urgency, flatulence with discharge, fat-soluble vitamin deficiency (take multivit 2 hours apart).
Contraindications
- Chronic malabsorption syndrome, cholestasis, pregnancy, lactation, on cyclosporin (absorption reduced).
