Mechanism of Action
Amoxicillin is a beta-lactam antibiotic that binds penicillin-binding proteins (PBPs) on bacterial cell walls. This inhibits the final transpeptidation step of peptidoglycan synthesis, weakening the cell wall and causing osmotic lysis of dividing bacteria. The aminopenicillin side-chain extends activity to many gram-negative organisms beyond classical penicillin spectrum.
Dosage & Administration
Children: 20–40 mg/kg/day in 3 divided doses, dispersed in water. Adults: 250–500 mg three times daily (every 8 hours). Course duration typically 5–7 days; longer for severe infections. Take with or without food.
Side Effects
Diarrhoea, nausea, vomiting, mild skin rash. Less commonly: pseudomembranous colitis with prolonged use, allergic skin reactions (urticaria, rash). Rare: anaphylaxis, hepatitis, Stevens-Johnson syndrome, interstitial nephritis.

Hypersensitivity to penicillins, cephalosporins or any beta-lactam antibiotic. History of severe allergic reaction to amoxicillin. Infectious mononucleosis (high rate of rash). Caution in renal impairment.
