About Lunomac-10 Tablet
Lunomac-10 Tablet contains Leflunomide 10 mg — the lower maintenance/starting dose DMARD for rheumatoid arthritis and psoriatic arthritis. Alternative to Methotrexate for patients with intolerance or contraindications.
Used after initial Methotrexate inadequate response or in combination with Methotrexate for severe RA.

Mechanism of Action
Leflunomide is an isoxazole DMARD — its active metabolite teriflunomide reversibly inhibits dihydroorotate dehydrogenase (DHODH), reducing pyrimidine synthesis. This selectively affects rapidly dividing lymphocytes — reducing T-cell and B-cell proliferation. Slower-onset (6–8 weeks) but disease-modifying in rheumatoid arthritis.
Dosage & Administration
Adults: 10 mg once daily as maintenance. May load with 100 mg/day for 3 days then 20 mg/day, then maintenance.
Side Effects
Diarrhoea, alopecia, rash, transaminase elevation (monitor LFTs), hypertension, hypersensitivity. Teratogenic — washout (cholestyramine) needed before pregnancy planning.
Contraindications & Precautions
Pregnancy, lactation, severe hepatic impairment, severe immunodeficiency, active infection, severe renal impairment, hypoproteinaemia.
