About Bacofer-10 Tablet

Bacofer-10 Tablet contains Baclofen 10 mg, a centrally acting skeletal muscle relaxant used for the treatment of spasticity in multiple sclerosis, spinal cord injury, cerebral palsy, motor neuron disease, stroke and traumatic brain injury. The 10 mg strength is the standard titration dose.

Baclofen is a structural analogue of GABA that selectively activates GABA-B receptors in the spinal cord, reducing the release of excitatory neurotransmitters at mono- and polysynaptic reflex pathways. This produces a reduction in muscle tone, painful spasms, clonus and flexor spasms — improving function and quality of life in patients with central spasticity.

Mechanism of Action

Baclofen is a selective GABA-B receptor agonist that hyperpolarises spinal interneurons and inhibits the release of excitatory neurotransmitters (glutamate, aspartate) at mono- and polysynaptic reflex arcs. This reduces gamma motor neuron activity and lowers muscle tone, alleviating spasticity without significantly affecting voluntary movement at therapeutic doses.

Dosage & Administration

Adults: start 5 mg three times daily, titrate every 3 days by 5 mg per dose up to 30 mg three times daily (max 80 mg/day). Reduce dose in renal impairment. Discontinue gradually — abrupt withdrawal may precipitate seizures, hallucinations or rebound spasticity.

Side Effects

Drowsiness, dizziness, nausea, weakness, fatigue, dry mouth, urinary frequency. Confusion in elderly. Constipation. Rare: respiratory depression, seizures, hepatitis, severe withdrawal syndrome on abrupt cessation.

Contraindications & Precautions

Hypersensitivity to baclofen. Active peptic ulcer. Severe renal impairment (CrCl < 30 — dose reduction; <10 — avoid). History of seizure disorder (caution). Caution in elderly and patients with respiratory disease. Avoid alcohol and sedatives.
