About Amivate-10 Tablet
Amivate-10 Tablet contains Amitriptyline Hydrochloride 10 mg, a classical tricyclic antidepressant (TCA) used across multiple specialties. While newer antidepressants have largely replaced TCAs for depression, Amitriptyline remains a first-line agent for neuropathic pain, fibromyalgia, chronic tension-type headache and migraine prophylaxis at low doses.
Amitriptyline blocks the reuptake of serotonin and noradrenaline at the presynaptic neuronal membrane, increasing synaptic monoamine availability. In neuropathic pain states this strengthens descending pain-inhibitory pathways from the brainstem, reducing the abnormal pain signals generated by damaged peripheral nerves. Mechanism of Action

Side Effects
Dry mouth, drowsiness (often desired at bedtime), constipation, blurred vision, urinary retention, weight gain, orthostatic hypotension. Important: cardiac conduction effects at high doses, particularly QT prolongation; ECG monitoring may be required in cardiac patients.

Hypersensitivity to TCAs. Recent myocardial infarction. Concomitant MAOI use (within 14 days). Severe hepatic impairment. Narrow-angle glaucoma. Acute mania. Caution in elderly, cardiac arrhythmias, prostate enlargement and epilepsy.
