Mechanism of Action
Clobetasol is a fluorinated glucocorticoid with very high glucocorticoid receptor affinity. It potently suppresses inflammatory gene transcription, immune cell infiltration and inflammatory cytokine production in the skin. The ultra-high potency comes at the cost of significantly higher systemic absorption and local atrophy risks compared with less potent steroids.
Dosage & Administration
- Apply a thin layer to affected area once or twice daily.
- Maximum 2 weeks at a time, maximum 50 g per week.
- Do not use under occlusion.
- Avoid face, groin and flexures.
Side Effects
- Skin atrophy, telangiectasia, striae, perioral dermatitis, acneiform eruptions, hypertrichosis.
- Systemic absorption can cause HPA-axis suppression (especially with prolonged use or large surface area).
- Tinea incognito if used on undiagnosed fungal infection.
Contraindications
- Hypersensitivity.
- Viral skin infections (herpes simplex, varicella).
- Bacterial or fungal skin infections (untreated).
- Rosacea, acne, perioral dermatitis.
- Use on face/groin/flexures (relative contraindication).
- Children (limit duration and area).
