About Amanzil-100 Tablet

Amanzil-100 Tablet contains Amantadine Hydrochloride 100 mg, a unique molecule that combines dopaminergic, NMDA-receptor antagonist and antiviral activity. It is used principally as adjunctive therapy in Parkinson’s disease — alone in early disease or combined with levodopa in advanced disease, where it reduces levodopa-induced dyskinesia.

Amantadine increases the release of dopamine from intact nigrostriatal terminals, inhibits dopamine reuptake and weakly antagonises NMDA glutamate receptors. The NMDA action is particularly valuable in suppressing levodopa-induced peak-dose dyskinesia, a major late-stage complication of Parkinson’s therapy. The drug also has historical activity against influenza A virus by blocking the viral M2 ion channel (though resistance is now near-universal).

Amanzil-100 is manufactured by Trumac Healthcare under WHO-GMP conditions and is available for monopoly-based PCD pharma franchise and third-party manufacturing — a steady-volume neurology SKU with strong specialist demand.

Indications

  • Idiopathic Parkinson's disease (monotherapy in early disease)
  • Levodopa-induced dyskinesia in advanced Parkinson's disease
  • Post-encephalitic and drug-induced parkinsonism
  • Multiple sclerosis-related fatigue (off-label specialist use)

Mechanism of Action

Amantadine enhances dopaminergic transmission by promoting presynaptic dopamine release, blocking dopamine reuptake, and weakly stimulating dopamine receptors. It is a non-competitive antagonist of NMDA glutamate receptors, which underlies its antidyskinetic effect by modulating overactive corticostriatal glutamatergic signalling.

Dosage & Administration

Adults: 100 mg twice daily; titrate to clinical response. Elderly and renally impaired patients: 100 mg once daily or less. Avoid late-evening doses (may cause insomnia). To be prescribed by a neurologist or physician.

Side Effects

Insomnia, nervousness, dizziness, peripheral oedema, livedo reticularis (mottled skin discolouration), dry mouth, blurred vision, constipation, urinary retention. Rarely: confusion, hallucinations (more common in elderly and renal impairment), seizures.

Contraindications & Precautions

Hypersensitivity to amantadine. Untreated narrow-angle glaucoma. Severe renal impairment. History of seizure disorder requires caution. Avoid abrupt withdrawal (risk of neuroleptic malignant syndrome-like reactions). Caution in elderly and patients with psychiatric history.

Storage

Store below 30 °C in a dry place, protected from light and moisture.
