Mechanism of Action
Domperidone is a peripheral D2 receptor antagonist — enhances upper GI motility, accelerates gastric emptying, increases LOS tone. Does not cross blood-brain barrier — no extrapyramidal effects at therapeutic doses.
Dosage & Administration
Infants: 0.25–0.5 mg/kg per dose, 3 times daily before feeds.
Side Effects
Hyperprolactinaemia (galactorrhoea, gynaecomastia, amenorrhoea), QT prolongation (short-term high-dose), GI cramps.
Contraindications & Precautions
Prolactinoma, QT prolongation, on QT-prolonging drugs, severe hepatic impairment, GI obstruction/haemorrhage.
